Key testing data of the BloodoTM NAD Test
1. Calibration and Error margin
NAD is measured according to a calibration curve received against the NAD standard (Figure 1). A typical calibration curve is conducted using NAD concentration range of 0-60 µmol/L (accounting for dilution),
enabling the determination of NAD in DBS samples within this range. The LOD of the BloodoTM NAD Test is 0.23 µmol/L and LOQ is 0.40 µmol/L for DBS samples (whole human blood). Multiple DBS samples were analyzed and the relative standard deviation was calculated. The data yielded a 6.6% relative standard deviation.
2. BloodoTM NAD Test validation data
The BloodoTM NAD test was validated by comparing the assay results between liquid and DBS samples (R2=0.94) (Figure 2). The comparison demonstrated excellent correlation, indicating that the DBS samples align closely with the liquid blood samples. To validate that only NAD is being assayed, it was selectively removed from samples using NAD-specific enzymes (Figure 3). Normal samples showed typical NAD levels, while samples with NAD removed had significantly lower levels, confirming the BloodoTM NAD Test's specificity.
3. NAD supplement effectiveness monitoring using BloodoTM NAD Test
BloodoTM NAD Test was used to measure NAD levels from DBS samples taken from volunteers who were using NAD supplementation. Initially, all volunteers provided DBS samples for 5 days without any supplementation in order to establish baseline NAD levels in their blood. All samples were collected first thing in the morning before any food or drink intake. From the 5th day, three volunteers took 1 g per day of nicotinamide mononucleotide (NMN) in the morning, while the fourth volunteer, serving as a control, took 1 g per day of nicotinamide. The results are given in Figure 4 and show that BloodoTM NAD Test measures NAD level changes in the blood after using NAD supplements.
